I am adding 301 redirect to htaccess file but I don't see it runs. My question is, how long for 301 redirect to take effect? Does this affect by web caching plugin that I am using on my website?

You should clear the cache after each update for your .htaccess file. Or you can try of using private/incognito browser mode to check for how the website is being loaded without cache.

In general, the rules provided in .htaccess file should take effect immediately. If you make adjustments on .htaccess and there are no effect on your website even after clearing cache, you should check your webserver's configuration, since there can be multiple .htaccess files or that it's usage can be disabled.

Agree with @hostens and I think your redirection codes are wrong thus it didn't take effects. I faced same problem and If possible, you should use PHP redirection code, it will work better.

If your site is using Wordpress, I would recommend Redirection plygin by John Godley, it works as a charm.

Sounds like a caching issue. 301 redirects work the moment you hit save.

It should be instant. You should clear the cache, delete the browsers cookies and also flush DNS.
